3T3 murine embryonic fibroblasts are used as feeder layers to support the proliferative potential of human primary keratinocytes during in vitro expansion for cell and gene therapy applications. To identify signals mediating this function, we analyzed fibroblast protein content using mass spectrometry. We compared the proteome of lethally irradiated fibroblasts used as feeder layers at two time points: 24 hours and 4 days post-seeding. Our findings highlight differences in extracellular matrix components between the two conditions.
