The Transversal–Longitudinal Pathogenetic Theory (TLPT) is a systems‑biology framework that models disease emergence, progression, and resolution through the interaction of master‑switch nodes, transversal fluxes, longitudinal architectural layers, and meta‑attractor dynamics. TLPT integrates causal chains, pathogenic axes, transversal flows, feedback loops, pathological attractors, and the transversal meta‑attractor into a unified pathogenetic pyramid composed of seven hierarchical levels. The theory defines explicit activation/deactivation laws for master‑switch nodes, eligibility rules for intervention modules, and criteria for transversal–longitudinal blockade and protective meta‑attractor formation. TLPT provides a mechanistic, network‑based interpretation of multisystemic pathology and resilience, enabling conceptual modeling of complex biological states without requiring experimental datasets or SBML formalization.This FAIRDOMHub project provides a structured, openly accessible repository for the TLPT framework, including the full theoretical manuscript, conceptual diagrams, module definitions, and transversal–longitudinal rules. The goal is to offer the systems‑biology community a coherent pathogenetic architecture that can be referenced, reused, extended, or integrated into modelling workflows, without requiring experimental datasets or computational implementations. TLPT is published here in its original form to ensure transparency, reproducibility, and long‑term accessibility.
