| summary | We aimed at indentifying genes involved in mealybug sex pheromone biosynthesis. As these should be expressed in virgin females and are presumably downregulated after mating, we looked at genes differentially expressed between mated and virgin females. | ||||||||

| rearing protocol | The colony of Planococcus citri was established at the Universitat Politècnica de València (UPV, Valencia, Spain) using starting specimens from Servicio de Sanitat Vegetal (Generalitat Valenciana, Silla (Valencia, Spain)). Mealybugs were reared on organic green lemons. These were previously covered with paraffin around the midsection to delay their desiccation. Insects were maintained in a rearing chamber (darkness, at 24 ± 2 °C and 40−60% relative humidity) in plastic boxes. | ||||||||
| treatment protocol | Mated females were collected from the main P. citri stock colony described in the rearing protocol. These were collected after checking for the presence of ovisacs with fertilized eggs, which means that females were effectively mated. Virgin females were obtained as described in Vacas et al. (2019). New lemons infested with P. citri first instars from the main stock colony were maintained in differentplastic boxes. After the second instar stage, males produce a cottony cocoon that covers their bodies to pupate. Then, those lemons were periodically checked for the presence of cocoons, which were manually removed with an entomological needle to avoid matings and leave lemons infested only with virgin females. | ||||||||

| Mealybug Stock Colony. The colony of D. aberiae was established n our facilities at the Universitat Politècnica de València (UPV, Valencia, Spain) using specimens from the Laboratory of Entomology, based at the Instituto Valenciano de Investigaciones Agrarias (IVIA, Valencia, Spain). Mealybugs were reared on organic green lemons, which were previously covered with paraffin around the midsection to delay their desiccation. Starting from gravid females and ovisacs deposited on lemons, newly hatched individuals establish on the surface of fruit and follow the developmental cycle. Mated females were allowed to oviposit. When ovisacs were laid, they were gently transferred with an entomological needle to new lemons. Insects were maintained in a rearing chamber, in the dark at 24 ± 2 °C, with 40−60% relative humidity. After the second instar stage, males produce a distinguishable cottony cocoon that covers their bodies to pupate and transform into winged adults. Lemons were visually inspected every 3−4 days for the presence of cocoons, which were manually removed with an entomological needle to leave lemons infested only with virgin females for volatile collection and profiling purposes. Mated females were sampled on the lemons from the main stock colony after checking for the presence of ovisacs. | |||||||||
