Methodology | ||||||||||||||||
The activity/pH profile was determined by incubating the enzyme at room temperature | ||||||||||||||||
in 50 mM of KPi pH=8, CHES pH= 9, CHES pH =10. All buffers contained 0.1 mM PLP. | ||||||||||||||||
Samples (90 μL) were taken from the reaction mixture at different time points and transfered to a 96-well plate. | ||||||||||||||||
The enzyme activity was photometrically determined according to the acetophenone assay (Schätzle et al. 2009). | ||||||||||||||||
Enantiopure (R)-phenylethylamine (R-PEA)/DMSO 1mM and Sodium pyruvate 4mM were then added | ||||||||||||||||
and the rate was determined by measuring the absorbance (245 nm) of acetophenone that is produced. | ||||||||||||||||
One unit of activity was defined as the amount of enzyme that produced 1 µmol acetophenone per minute. | ||||||||||||||||
Assay Conditions | ||||||||||||||||
Reaction mixture: 100 μL purified enzyme + 1,7 mL buffer | ||||||||||||||||
Incubation and measurements were done at room temperature. | ||||||||||||||||

CHES 50 mM/ PLP 0.1 mM pH = 9 | ||||||||||||||||

The assay componets were pipetted in the appropriate well in the following order: | ||||||||||||||||
10 μL of amine stock solution (20 mM R-PEA/DMSO) | ||||||||||||||||
90 μL reaction mixture | ||||||||||||||||
100 μl of pyruvate stock solution (4 mM, ddH2O) | ||||||||||||||||
