Description

LUC imaging under 3:21 cycles then constant light. Replicate of experiment AT0064; similar to AT0007, AT0028.

Purpose

photoperiod alteration study

Comments

Imaging started 23/01/07. Fungal infection on GI and CCR2 cca1 lhy mutant plate. CAB, GI, CCA1, CCR2 cca1 lhy groups badly affected. TOC1 data show anomalous dawn peak, so TOC1 data not used for phase analysis in Edwards et al. 2010 Curated by Andrew Millar, 2013.

Sample preparation

Samples were sprayed with luciferin once, upon transfer from entrainment to measurement conditions

imaging script starts 5 minutes before required ZT, then 55 minutes interval, 5 minutes light off, 30 minutes capture

Growth on Agar 1.2% MS 0.5;Agar 1.5% MS 1 3% Sucrose for 6.0 days (LD12:12 22c). entrainment

Growth on Agar 1.5% MS 1 3% Sucrose for 7.0 days (4x3:21-LL). first day not imaging, 4 days 3:21, 3 days constant red+blue light. P13H1 = Percival chamber number 13, Hamamatsu camera 1.
