LUC imaging 12:12 LD to LL in RB light

Purpose

testing different photoperiod effect on entrainment and phase on release to LL

Comments

CCR2:LUC in cca1 lhy and in cca1lhygi had white fungal contamination all over plate ELF3:LUC had low germination

Sample preparation

sprayed upon transfer from entrainment

Luciferase luminescence (HAM 1)

imaging script starts 5 minutes before required ZT, then 55 minutes interval, 5 minutes light off, 30 minutes capture

Experimental conditions

LD12:12 22c

Growth on Agar 1.5% MS 1 3% Sucrose for 6.0 days (LD12:12 22c). entrainment
